Biography

L'attività di ricerca del Dr. Lasorsa riguarda l'identificazione funzionale di nuovi geni codificanti carrier mitocondriali e il loro coinvolgimento nel metabolismo cellulare e in malattie nell'uomo. I carriers mitocondriali rappresentano la più importante via di comunicazione metabolica tra la matrice mitocondriale e il citosol catalizzando il trasporto specifico di un vasto numero di metaboliti attraverso la membrana interna dei mitocondri. Mediante lo sviluppo di tecnologie recombinanti per l'espressione e la ricostituzione funzionale dei carriers, nella sua carriera scientifica il Dr. Lasorsa ha contribuito all'identificazione di cinque geni di lievito e quattro geni umani codificanti nuovi carriers mitocondriali insieme con altri geni di carrier mitocondriali di organismi intermedi. La strategia di ricerca impiegata consiste nell'over-espressione in E. coli o S. cerevisiae dei geni codificanti i carrier mitocondriali, purificazione delle proteine e la loro ricostituzione funzionale in liposomi. Il Dr Lasorsa ha anche maturato esperienze in laboratori internazionali che hanno permesso l'acquisizione e lo sviluppo di nuove tecniche sperimentali per la caratterizzazione del ruolo fisiologico dei carriers mitocondriali in lievito e in cellule di mammifero. Il Dr Lasorsa ha anche contribuito all'identificazione dei meccanismi patogenici di malattie metaboliche recentemente associate a disfunzioni di carrier mitocondriali quali la CTLN2 (OMIM,603471) e la AGC1 deficiency (OMIM, 612949). The scientific research of F.M.Lasorsa deals with the identification of new genes encoding mitochondrial carriers and their involvement in cell metabolism and human diseases. Mitochondrial carriers comprise a family of 40 to 50 proteins depending on the organism and provide the principal routes of communication between the mitochondrial matrix and the extra-mitochondrial spaces by transporting a wide range of metabolites, nucleotides and cofactors. By developing recombinant technologies for the expression and functional reconstitution of carriers, F.M.L. contributed to the identification and biochemical characterization of several genes encoding new mitochondrial carriers. The employed experimental platform consists of gene over-expression in E. coli or S. cerevisiae, purification of proteins and their functional reconstitution in liposomes. F.M.L. during his scientific career also matured experiences in other laboratories in Italy and abroad thus acquiring and developing new techniques aimed at the characterization of the physiological roles of mitochondrial carriers in yeast as well as in mammalian cell models. F.M.L. also contributed to the identification and elucidation of the pathogenesis of metabolic diseases associated with mitochondrial carriers, such as type-II citrullinemia, brain AGC1 deficiency, GC1 deficiency and PNC2 deficiency.
